Purpose
PT-141 (bremelanotide) is the reference MC4R-selective melanocortin agonist studied in sexual-health research. Unlike its non-selective precursor melanotan-II, PT-141 minimises the cutaneous-pigmentation and appetite-suppression effects associated with broad melanocortin agonism while preserving the central-nervous-system pathway most closely tied to sexual-arousal biology.
This stack treats PT-141 as a monostack for its intended research use. Melanotan-II is included in the reference set for mechanistic comparison but is not administered concurrently in reported research protocols.
Component peptides
- PT-141 — 1,000–2,000 mcg subcutaneously on an as-needed basis, with the reference protocol limiting to one dose per 24 hours and no more than eight doses per month.
- Melanotan-II — included for reference only; not administered concurrently with PT-141 in reported research writeups.
Timing & rotation
The reference research protocol calls for administration approximately 45 minutes before anticipated activity. Chronic daily dosing is not described in the primary clinical literature.

- PT-141 10 mg vial in 2 mL BAC → 5,000 mcg/mL; 1,750 mcg dose = 0.35 mL = 35 units.

Reported outcomes
Phase-3 trial data (RECONNECT) reported improvements versus placebo on desire and distress domain scores. Male erectile-function trial data reported earlier are older and less rigorously controlled.
